Cluster of differentiation 28 (CD28) is a crucial receptor expressed on the surface of T cells and plays a pivotal role in the regulation of the immune system. The engagement of CD28 with its ligands expressed on antigen-presenting cells (APCs) provides a co-stimulatory signal that is essential for T cell activation, proliferation and production of interleukin-2 (IL-2). Moreover, CD28 activation contributes to the development of memory T cells, which confer sustained immunity. Dysregulation of CD28 results in various autoimmune diseases, while its modulation holds therapeutic promise, especially in cancer immunotherapy.
